NIEHS In Vivo Repeat Dose Biological Potency Studies

The National Institute of Environmental Health Sciences (NIEHS) Division of Translational Toxicology (DTT) conducts in vivo repeat dose biological potency studies to estimate a chemical's potency by analyzing transcriptomic changes from short-term animal studies using standardized protocols. Similar to the EPA's Transcriptomic Assessment Product (ETAP), these NIEHS reports use mathematical model-based approaches to identify and report the potency of dose-responsive effects without making subjective interpretations or hazard conclusions. The methodology is based on a peer-reviewed and approved standardized process, where the study design, analysis, and results presentation are consistently applied across different chemicals. This approach is founded on scientific evidence demonstrating that the transcriptomic potency derived from a short-term (e.g., 5-day) in vivo study can serve as a reasonable approximation of the toxicological potency observed in more resource-intensive, long-term guideline assessments.
